Nirmatrelvir And Ritonavir and Covid-19: is the FAERS signal real?

n=— co-reports · PRR — · ROR —
A written Bradford Hill assessment of one drug–event pair. The grade is a judgement about the signal, not a statement that this drug causes this event in any one person; FAERS counts are reports, not rates.

Nirmatrelvir And Ritonavir ↔ Covid-19

n=— co-reports · PRR — · ROR —

Nirmatrelvir/ritonavir (Paxlovid) is an oral antiviral for mild-to-moderate COVID-19. Co-reporting of COVID-19 as a reaction is partly a coding artifact (indication overflow) and partly the viral rebound phenomenon. 2,588 co-reports (PRR 62.2) reflect the treated population and known rebound, not causation.

Coding artifact / rebound. Paxlovid label (no boxed warning for COVID-19). Rebound: NEJM 2022, PMID: 37596423.

✗ No ClinicalTrials.gov results-posting trial in the top search hits for Nirmatrelvir captured covid-19 as a tracked adverse event in a usable structure. The EPIC trial (NCT04960203) tracked COVID-19-related AEs; rebound is well-described.
